Her mother, Christine worked as a clothing store manager and her father, Greig Johnson worked as a salesman at an automobile company. Amy was raised alongside 2 older siblings, a brother named Grieg Johnson Jr. and a sister named Julie Johnson Clary. However, her mother died on August 19, 1998, after a long battle with cancer.

Johnson had her high school education at Denis-Yarmouth Regional High School located in Yarmouth, Massachusetts. She was a very active teenager as she participated in gymnastics. As a high schooler, Johnson took part in several gymnastics events in the United States and beyond. She equally took interest in acting as she appeared in several local theatre productions. Amy Jo Johnson eventually quit gymnastics at the age of 17 in order to pursue a full-time career in acting.
