Dejounte Murray began his professional basketball career in March 2016, after being selected with the 29th overall pick in the 2016 NBA draft by the San Antonio Spurs. He played in the 2016 NBA Summer League, after signing a rookie contract with the club. He made his debut for the team on October 26 where he recorded 1 assist and 2 rebounds in a 98–79 victorious win against the New Orleans Pelicans. He made a career high of 24 points in a 118–104 win against the Denver Nuggets, making him the youngest player in Spurs history to do so, breaking the record of . He was also assigned to go to the Austin Spurs of the NBA Development League during his first season.

In his second season in the club, Dejounte Murray started in the season’s opening game in October 2017 and recorded 16 points, 5 rebounds and 2 assists in a 107–99 win against the Minnesota Timberwolves. In the 2017/2018 season, he became the first player since with 500 points and 300 rebounds in his first 100 games with the Spurs.
