He was born on April 18, 1934, in New York to parents; James Child Drury his father, and Beatrice Crawford Drury, his mother. James Dury spent a greater part of his childhood in New York. Always inclined to arts, he opted to study drama at New York University. James started his career at a very young age, at the age of 12, he appeared on Life with Father. He was first signed with MGM and later on 20th Century Fox. The latter deal saw the New Yorker star in Love Me Tender (1956). He also starred in the movie Bernardine in 1957.

The talented actor went on to appear in lots of TV series including Richard Diamond, Private Detective, Black Saddle, Death Valley Days, The Rebel and many others. In the early 60s, he made various secondary appearances for Walt Disney. James Drury went on to star in the Western film Ride the High Country. Afterward, he landed a leading role on the popular The Virginian series. Drury starred as the ranch foreman in the notable series which ran from 1962 to 1971. He also reprised his role in The Men from Shiloh, a sequel to The Virginian.

Drury greeted the 90s with a remarkable performance in the first episodes of the series Walker, Texas Ranger. The Virginian made a return to the screens in 2000 but as a TV movie. James as the lead character in the series made a cameo appearance in the movie. He is known for so many TV series of different genres and was remarkable for his roles.
