Her place of birth was in Manhattan, New York City, the United States which makes her an American citizen and she belongs to the White ethnicity. Spiro also spent her formative years in New York where she completed her high school education before proceeding to take a course at Circle in the Square Theatre School. From there, she went further to the Royal Academy of Dramatic Art. She later delved into filmmaking and pursued the MFA Program thereof, earning a degree from the Columbia University.

Jordana Spiro then commenced her acting career playing minor roles in television series and films in 1995. Some of her early appearances can be seen in films like If These Walls Could Talk where she played the character of Alison and this was followed with her role of Freddie in Silk Stalking. Spiro made an outstanding appearance in 2001 when she played the recurring role of Brandi in The Huntress and that same year, she was in the popular CSI: NY where she depicted the character of Tavia Greenberg.
