Mary Rose Byrne was born on July 24, 1979, in Balmain, a suburb of Sydney, New South Wales. She has Scottish and Irish ancestry. Her mother Jane worked as the administrator of an elementary school while her father Robin Byrne served as a statistician. The youngest of her parents’ 4 children, Rose Byrne was raised alongside sisters Alice and Lucy and a brother named George.

Byrne had her early education at Balmain Public School. As an 8-year old, she was enrolled for acting classes and later became a member of the Australian Theatre for Young People. For high school, Byrne attended Hunters Hill High School and Bradfield Senior College. She had her college education at the University of Sydney and later studied acting at the Atlantic Theater Company in Manhattan, New York City.
